ABSTRACT

El uso de biomarcadores hematológicos es importante para poder evaluar riesgos ambientales y la exposición a sustancias químicas potencialmente tóxicas y así poder desarrollar medidas que sirvan como señales de alarma temprana en ambientes contaminados. El objetivo de este trabajo fue determinar biomarcadores hematológicos en el sapo común Bufo (Rhinella) arenarum que proporcionen información para el diagnóstico de la salud de distintos ecosistemas de la provincia de Córdoba. Un total de 93 individuos adultos de Bufo (Rhinella) arenarum fueron capturados en cuatro sitios de la provincia de Córdoba y distintas variables ambientales del agua fueron medidas en cada sitio. Las muestras de sangre se obtuvieron mediante la punción de la vena angularis. Luego fueron teñidas con la tinción diferencial May Grünwald-Giemsa y fueron observadas mediante microscopio Zeiss Primo Star iLED para realizar el conteo de glóbulos blancos. Las variables ambientales del agua mostraron diferencias estadísticamente significativas entre las poblaciones, se registraron valores menores en “Alpa Corral”, mientras que “Villa Dálcar” presentó los mayores valores para dichas variables. No se encontraron diferencias estadísticamente significativas entre localidades en el recuento de glóbulos blancos (cada 1000 eritrocitos); sin embargo, en “Villa Dálcar” se registraron los ejemplares con mayor cantidad de leucocitos en el conteo de glóbulos blancos (425,48 ± 481,34). Mientras que para la fórmula leucocitaria, los linfocitos fueron las células más abundantes para los ejemplares de todos los sitios; encontrándose diferencias estadísticamente significativas entre cada localidad y “Alpa Corral”. Los ejemplares de “Villa Dálcar” presentaron diferencias estadísticamente significativas en relación a los monocitos y basófilos, registrándose altos valores para ambas células en los ejemplares de este sitio; mientras que en los ejemplares de “Cultivo” las diferencias se encontraron en eosinófilos y la relación neutrófilos/linfocitos. Estos resultados revelaron que de las cuatro localidades analizadas, “Villa Dálcar” representa la más afectada por las actividades antrópicas. Estas variaciones registradas en los parámetros hematológicos podrían considerarse evidencia de la posible presencia de agentes contaminantes en el lago “Villa Dálcar”.


Hematologic biomarkers are important to assess the environmental and health risks of exposure to potentially toxic chemicals, and for developing measures that serve as early warning signals in polluted areas. The aim of this study was to determine hematologic biomarkers of common toad Bufo (Rhinella) arenarum that provide information for the diagnosis of the health of ecosystems of Córdoba province. A total of 93 adult individuals of Bufo (Rhinella) arenarum were collected from four sites of Córdoba Province and hydric environmental variables were measured on each site. Blood samples were obtained by angularis vein puncture, smears of fresh blood were stained with May Grünwald-Giemsa and observed by using a microscope Carl Zeiss Primo Star (Pack 5), for white blood cell count. Water environmental variables showed statistically significant differences among localities, with the lowest values in “Alpa Corral” and highest values in “Villa Dálcar”. There were no statistically significant differences among individuals of different sites for white blood cells count. However, individuals from "Villa Dálcar" showed the highest number of white cells (425.48 ± 481.34). In the leukocyte formula, lymphocytes were most abundant white blood cells in individuals from all study sites. The blood parameters of toads from all sites showed statistically significant differences with individuals from “Alpa Corral”. "Villa Dálcar" Individuals exhibited significant differences in relation to monocytes and basophils with the highest values for both cell types. “Cultivo” locality showed significant differences for eosinophils and for the relationship neutrophils/ lymphocytes. These results revealed that “Villa Dálcar” represent the site most affected by human activities. In particular, changes recorded in hematological parameters could be giving evidence of the possible presence of contaminants in Lake “Villa Dálcar”.

ABSTRACT

Los anfibios son constituyentes integrales de ecosistemas naturales y agrícolas, y debido a su sensibilidad pueden ser utilizados como bioindicadores. La frecuencia de micronúcleos se emplea como un biomarcador que evalúa la respuesta a agentes genotóxicos. El objetivo del trabajo fue determinar y comparar la frecuencia de micronúcleos en eritrocitos de Bufo arenarum que habitan ambientes antrópicos de la ciudad de Río Cuarto. Se escogieron tres sitios modificados: un lago urbano Lago Villa Dálcar (33º06´S-64º22´W) y dos sitios de cultivo denominados Cultivo1 (33º05'S-64º26'W) y Cultivo 2 (33º05'S-64º25'W). Como control se escogió un área ubicada en las sierras de Comechingones, en el Campo Experimental Las Guindas (32º35'S- 64º42'W), pertenecientes a la UNRC. En cada sitio se recolectaron ocho individuos utilizando trampas de caída y relevamientos por encuentros visuales. A cada individuo se le extrajo sangre de la vena angularis inmediatamente después de la captura. Se realizaron dos extendidos por cada individuo, se dejaron secar al aire por 24 hs, luego se fijaron con metanol por 20 minutos y se colorearon con Giemsa al 5% durante 10 minutos. Posteriormente los frotis fueron analizados con microscopio (1000X) y se tomaron fotografías con cámara digital. La frecuencia de micronúcleos fue contabilizada para 4000 células por individuo. Se realizaron comparaciones utilizando el test de Kruskal-Wallis. Las frecuencias de micronúcleos encontradas son: Las Guindas: 0,17 ± 0,23; Cultivo1: 0,78 ± 0,28; Cultivo 2: 0,61 ± 0,45 y Villa Dálcar: 0,76 ± 0,47. Existen diferencias significativas entre el control y los demás sitios (p< 0,01). Los resultados obtenidos demuestran la relevancia en la aplicación del test de micronúcleos in vivo en anfibios anuros para comparar distintos ambientes. Estos resultados indicarían la necesidad de nuevas investigaciones y de un monitoreo más prolongado en el tiempo para estos ambientes, incluyendo por ejemplo otros tipos de biomarcadores, así como otras posibles especies representativas de la región.


Amphibians are integral constituents of natural and agricultural ecosystems, and because of their sensitivity can be used as bioindicators. Micronuclei test is used as a biomarker for determining the response to genotoxic agents. The purpose of this study was to determine and compare the frequency of micronuclei in peripheral blood erythrocytes of Bufo arenarum toads that inhabiting anthropic environments for the city of Rio Cuarto. Organisms used in the MN test were collected from three sites modified: an urban lakeVilla Dalcar (33º06´S-64º22´W) and two cultivation sites called Crop 1 (33º05'S-64º26'W) and Crop 2 (33º05'S-64º25'W). As a control site, an area in the mountains of Comechingones, Las Guindas (32º35'S-64º42'W), belonging to the UNRC was sampled. At each site, eight individuals were collected using pitfall traps and by visual encounter surveys. To each individual is bled from the vein angularis immediately after capture. We performed two extended for each individual, air-dried for 24 hours, then fixed with methanol for 20 minutes and stained with 5% Giemsa for 10 minutes. Then the smears were analyzed with microscope (1000X) and took pictures with digital camera. For each individual the frequency of micronuclei for 4000 cells was recorded. Comparisons were made using the Kruskal-Wallis test. Micronuclei frequencies found are: Las Guindas: 0.17± 0.23; Crop 1: 0.78 ± 0.28, Crop 2: 0.61 ± 0.45 and Villa Dalcar: 0.76 ± 0.47. There are significant differences between the control and the other sites (p< 0.01). These findings show the relevance in the application of micronucleus test in vivo for anuran amphibians to compare different environments. These results suggest the need for further research and monitoring longer time for these environments, including such other biomarkers, as well as other possible species representative of the region.

Trophic ecology in tadpoles of Rhinella arenarum (Anura: Bufonidae) in agroecosystems and their possible implications for conservation. The progress of the agriculture border has led an important loss of natural habitats, with significant consequences for biodiversity. In this sense, the studies in anuran amphibian tadpoles inhabiting these environments are relevant, because the larval stage is a phase of population regulation. The aim of this study was to analyze the body condition and diet in Rhinella arenarum, tadpoles, an anuran species widely distributed in South America and that inhabit agroecosystems. Three sites were sampled, two agroecosystems with different alteration degrees (C1 and C2) and an uncultured (SM) third place. The captured tadpoles were anesthetized, fixed and preserved in formaldehyde (10%). Subsequently, body measurements were made and the complete intestine was removed and analyzed for food items under a binocular microscope. The diet in R. arenarum tadpoles has a dominance of algae Bacillariophyceae, followed by Cyanophyceae. In particular, the class Bacillariophyceae, due to the presence of the genus Navicula, Nitzschia, Gomphonema and Hantzschia, was important in the diet of the anurans in those agroecosystems. Class Cyanophyceae, mainly represented by genus Oscillatoria and Euglenophyceae represented by Euglena and Strombomonas, were predominant in the diet of the anurans in SM. Some differences in the total items consumed by tadpole were observed between the studied sites. Tadpoles that inhabit the modified sites (C1 and C2) recorded a significantly smaller amount of food. Moreover, the tadpoles that inhabit these sites showed a lower body condition. The presence of certain algae associated with eutrophic environments, could indicate some pollution in agroecosystems (C1 and C2). Food resources would be lesser in places with strong agricultural activity, possibly with a greater degree of eutrophication. A smaller food amount could have consequences at population level for the short and long time terms, because of its impact on individual growth. El crecimiento de la agricultura produce perdida del habitat natural, con consecuencias para la biodiversidad de los anfibios. Se analizo la dieta y condición corporal de larvas de anuros de Rhinella arenarum que habitan agroecosistemas. Los muestreos fueron realizados en dos agroecosistemas y en un tercer sitio, no afectado por cultivos. Las larvas capturadas fueron anestesiadas, fijadas y preservadas en formaldehido, se realizaron medidas corporales y se analizo la dieta de las larvas. Se registro diversidad en la oferta alimenticia proporcionada por los distintos ambientes. La dieta tuvo una predominancia de algas Bacillarophyceae, seguidas por Cyanophyceae. Particularmente, los géneros Navicula, Nitzschia, Hantzschia y Gomphonema (clase Bacillarophyceae), fueron importantes en los agroecosistemas. Los géneros Osillatoria, Euglena y Strombomonas (clases Cyanophyceae y Euglenophyceae), predominaron en la dieta de las larvas de anuros en el sitio menos alterado. Las larvas de los sitios más alterados consumen menor cantidad de alimento y registran una menor condición corporal. La presencia de determinadas algas, indicarían un mayor grado de contaminación en los agroecosistemas. La eutrofización de lagunas podría alterar la disponibilidad de alimento para larvas de anuros que podría tener consecuencias poblacionales negativas. Se sugiere el análisis de dietas larvarias como un potencial bioindicador de salubridad ambiental.

ABSTRACT

The regulation of body temperature in ectotherms has a major impact in their physiological and behavioral processes. Observing changes in thermal parameters related with reproduction allows us to better understand how Rhinella arenarum optimizes a thermal resource. The aim of this study was to compare the thermal parameters of this species between breeding and non-breeding periods. In the field, we recorded the body temperature from captured animals, the air temperature, and the temperature of the substrate. In the laboratory, we measured the temperature R. arenarum selected on a thermal gradient and the critical extreme temperatures. The results of our study show variations in thermal parameters between the two situations studied. This species makes efficient use of thermal resources during the breeding period by basking to significantly increase body temperature. Because calling is energetically costly for males, this behavior results in increased efficiency to callers during the breeding period. Rev. Biol. Trop. 59 (1): 347-353. Epub 2011 March 01.


La regulación de la temperatura en ectotérmos tiene gran importancia en los procesos fisiológicos y comportamentales. Los cambios en los parámetros térmicos relacionados con la reproducción nos permiten entender de qué manera Rhinella arenarum optimiza el recurso térmico. El objetivo del presente trabajo fue comparar los parámetros térmicos de la especie entre el periodo reproductivo y no reproductivo. En el campo se registraron la temperatura corporal de los animales capturados, la temperatura del aire y del sustrato. Además, en laboratorio se registro la temperatura selecta en un gradiente térmico. Como así también las temperaturas criticas máxima y mínima. Los resultados de nuestro estudio muestran variaciones de los parámetros térmicos entre ambas situaciones estudiadas. Aparentemente esta especie hace un uso eficiente del recurso térmico durante el periodo reproductivo ya que el basking le permite aumentar considerablemente la temperatura corporal, probablemente este comportamiento esté relacionado con el aumento de la eficacia para emitir el canto durante el periodo de reproducción ya que el mismo es energéticamente costoso para los machos.

Cell adhesion molecules act as signal transducers from the extracellular environment to the cytoskeleton and the nucleus and consequently induce changes in the expression pattern of structural proteins. In this study, we showed the effect of thyroid hormone (TH) inhibition and arrest of metamorphosis on the expression of E-cadherin, β-and α-catenin in the developing kidney of Bufo arenarum. Cell adhesion molecules have selective temporal and spatial expression during development suggesting a specific role in nephrogenesis. In order to study mechanisms controlling the expression of adhesion molecules during renal development, we blocked the B. arenarum metamorphosis with a goitrogenic substance that blocks TH synthesis. E-cadherin expression in the proximal tubules is independent of thyroid control. However, the blockage of TH synthesis causes up-regulation of E-cadherin in the collecting ducts, the distal tubules and the glomeruli. The expression of β-and α-catenin in the collecting ducts, the distal tubules, the glomeruli and the mesonephric mesenchyme is independent of TH. TH blockage causes up-regulation of β-and α-catenin in the proximal tubules. In contrast to E-cadherin, the expression of the desmosomal cadherin desmoglein 1 (Dsg-1) is absent in the control of the larvae kidney during metamorphosis and is expressed in some interstitial cells in the KClO4 treated larvae. According to this work, the Dsg-1 expression is down-regulated by TH. We demonstrated that the expression of E-cadherin, Dsg-1, β-catenin and α-catenin are differentially affected by TH levels, suggesting a hormone-dependent role of these proteins in the B. arenarum renal metamorphosis.


Moléculas de adesão celular atuam como tradutores do ambiente extracelular para o citoesqueleto e o núcleo e, conseqüentemente, induzindo mudanças no padrão da expressão das proteínas estruturais. Neste estudo, observamos os efeitos da inibição do hormônio tireóidea (TH) e detenção da metamorfose na expressão da E-caderina, β- e α- catenina no desenvolvimento do rim do Bufo arenarum. As moléculas de adesão celular durante o desenvolvimento têm uma expressão temporal e espacial seletiva, sugerindo um papel específico na nefrogênese. Com o propósito de estudar os mecanismos de controle da expressão das moléculas de adesão durante o desenvolvimento renal, bloqueou-se a metamorfose do B. arenarum com uma substancia goitrogênica que bloqueia a síntese de TH. A expressão da E-caderina nos tubos proximais é independente do controle da tireóide. Entretanto, o bloqueio da síntese de TH provoca uma sobre elevação da E-caderina nos dutos coletores, nos tubos distais e nos glomérulos. A expressão da β- e α-catenina nos dutos coletores, nos tubos distais, nos glomérulos e no mesênquima mesonéfrico é independente da TH. O bloqueio da TH causa uma sobre-regulação da β- e α-catenina nos tubos proximais. Em contraste com a E-caderina, a expressão da caderina desmossomal demogloína 1 (Dsg-1) é ausente no controle durante a metamorfose da fase larval dos rins e se expressa em algumas células intersticiais nas larvas tratadas com KClO4. De acordo com este trabalho, a expressão Dsg-1 é subregulada pela TH. Demonstramos que a expressão da E-caderina, Dsg-1, β-catenina e α-catenina são afetadas de forma diferencial pelos níveis de TH, sugerindo um dependência hormonal destas proteínas na metamorfose renal do B. arenarum.

ABSTRACT

There is evidence that environmentalmetal levels affect the immune function. In the particular case of the impact of heavy metals, information available suggests that the immune system is a target for low-dose Pb exposure. Among vertebrates it was shown that amphibians are capable of forming antibodies against a variety of antigens, causing several responses such as anaphylactic response and rejectinggrafts. In this study, the production of antibodies was assessed against sheep red blood cells (SRBC) in the anuran Bufo arenarum after six weekly injections of sublethal doses of lead (50 mg.kg-1, as lead acetate). Natural antibodies (natural heteroagglutinins)were also quantified against SRBC. Both assessments were carried out employing an ELISA method developedto this end, measuring absorbance (A). For natural anti-SRBC antibodies in both control (C) and Pb treated (T) toads, there was a non significant tendency to increase the initial absorbances (C initial: 0.69+0.39 A; T initial: 0.54+0.30 A), relative to those registered at the end of the experiments (C final: 0.89+0.49 A; T final: 0.76+0.31A); the T/C ratios also did not show changes. The only significant difference was found between initial and final samples from lead-treated toads (p<0.014). The immune anti-SRBC antibody levels of toads immunized with SRBC showed a significant lower increase (p<0.05) in lead-treated animals (T final: 0.66+0.36 A), as compared to control toads (C final: 0.91+0.50 A) at the end of the experiment. It was thus concluded thatthe changes due to the assayed doses of Pb in the levels of antibodies cannot be explained on the basis of only one singleaction mechanism of the metal, but as the result of a conjunction of effects over different immunocompetent cell subpopulations. These different responses suggest that factors affecting animals exposed to a foreign stimulus are different from those influencing the response of wild animals.


Existe evidencia de que los niveles de metal ambientales afectan la función inmune. En el caso particular del impacto de metales pesados, la información disponible sugiere que el sistema inmune es un blanco para la exposición a bajas dosis de Pb. Entre los vertebrados, se ha mostrado que los anfibios son capaces de formar anticuerpos contra una variedad de antígenos, que causan diversasrespuestas, tales como respuesta anafiláctica y rechazo de injertos. En este estudio, la producción de anticuerpos fue evaluada contra eritrocitos de oveja (EO) en el anuro Bufo arenarum, luego de seis inyecciones semanales de dosis subletales de plomo (50 mg.kg-1, como acetato de Pb). Los anticuerpos naturales (heteroaglutininas naturales) fueron también cuantificados contra EO. Ambas evaluaciones fueron llevadas a cabo empleando un método de ELISA desarrollado a este fin, midiendo laabsorbancia (A). Para los anticuerpos anti-EO naturales, tanto en sapos controles (C) como en sapos tratados con Pb (T), hubo una tendencia significativa a incrementar las absorbancias iniciales (C inicial: 0,69+0,39 A; T inicial: 0,54+0,30 A); la relación T/C tampoco mostró cambios. La única diferencia significativa se encontró entre las muestras inicial y final de los sapos tratados con plomo (p<0,014). Los niveles de anticuerpos anti-EO inmune de sapos inmunizados con EO mostraron un bajo incrementosignificativo (p<0,05) en los animales tratados con plomo (T final: 0,66+0,36 A), al compararse con sapos control (C final: 0,91+0,50 A) al final del experimento. De este modo, se concluye que los cambios debidos a las dosis analizadas de Pb en los niveles de anticuerpos no pueden explicarse sólo sobre la base de un único mecanismo de acción del metal, sino como resultadode una conjunción de efectos sobre diferentes subpoblaciones de células inmunocompetentes. Estas diferentes respuestas sugieren que los factores que afectan los animales expuestos a un estimulo externo son diferentes...

ABSTRACT

CAS might have a key role in the apoptosis induced by toxins, acting as anti-apoptotic factor, stimulating the cellular proliferation and the cell contact stabilization. To start to elucidate their role in the brain apoptosis of Bufo arenarum induced by cypermethrin (CY), the expression patterns of CAS and several cell adhesion molecules (CAMs) were established. Bufo arenarum tadpoles of the control and acute bioassay survival at different doses (39, 156, 625 and 2,500 microg CY/L) and times (24, 48, 72 and 96 h) of CY treatment were fixed in Carnoy, embedded in paraffin and sectioned. CAS and CAMs expression was determined by immunofluorescence and immunohistochemistry, respectively. When the bioassay starts, CAS increases suggesting a proliferative or regenerative effect, but decreases when the doses and/or the bbiocide exposure time increases, suggesting compromise of the cellular cycle control and trigger of an apoptotic wave. However, these neurotoxic mechanisms should not involve degradation of N-cadherin and alpha-catenin, in contrast of beta-catenin and axonal N-CAM180, at least in the initial apoptotic phase. Additionally, an adhesion compensatory mechanism by N-CAM180 is observed in the neuron cell body. These results suggest a dual role of CAS in the cellular cycle control during the CY-induced apoptosis: induction of cell proliferation and stabilization of the cell-cell junctions by modulating CAMs expression.

ABSTRACT

Amphibians respond to microbial infection through cellular and humoral defense mechanisms such as antimicrobial protein secretion. Most humoral defense proteins are synthetized in the skin. In this study we isolated two beta-galactoside-binding lectins with molecular weights of 50 and 56 KDa from the skin of Bufo arenarum. These lectins have significant hemagglutination activity against trypsinized rabbit erythrocytes, which was inhibited by galactose-containing saccharides. They are water-soluble and independent of the presence of calcium. The antimicrobial analysis for each lectin was performed. At mumolar concentration lectins show strong bacteriostatic activity against Gram negative bacteria (Escherichia coli K12 4100 and wild strains of Escherichia coli and Proteus morganii) and Gram positive bacteria (Enterococcus faecalis). The antibacterial activity of these lectins may provide an effective defense against invading microbes in the amphibian Bufo arenarum.

ABSTRACT

The ultrastructure of the intermediate segment (IS), distal tubule and collecting tubule (CT) of the south american toad Bufo arenarum, was studied by light and transmission electron microscopy. The IS is composed of cubical ciliated cells which propel the urine along the renal tubule. The distal tubule is divided into two portions: the early distal tubule (EDT) and the late distal tubule (LDT). The EDT is characterized by only one type of cells with well developed basolateral interdigitations and numerous elongated mitochondria, which are oriented normal to the basal surface. The "macula densa--like" is a specialized zone of the EDT in contact with the vascular pole, where cells are more tightly packed than in the rest of the tubule. The LDT shows two types of cells called dark and light cells according to the appearance of their cytoplasm. Dark cells have microplicae and few but long microvilli at their luminal surface, and abundant mitochondria in their cytoplasm. Light cells show basal and lateral infoldings and few mitochondria. The CT, which is composed of dark and light cells, exhibits an enlarged lumen with an undulated surface and dilated spaces between neighbouring cells. This work is a contribution to the knowledge of the kidney of B. arenarum; frequently used as an experimental model for physiological and biochemical studies.

ABSTRACT

Comprender los procesos que determinan la toxicidad de diversas sustancias a nivel celular constituye un desafío para el desarrollo tecnológico actual. Los ovocitos de diversas especies son utilizados con distintos propósitos en investigación. El tamaño de los ovocitos de anfibios y la facilidad con que puede obtenerse un gran número de ellos como células aisladas, hacen de los mismos el sistema de elección para múltiples propósitos, tales como: estudios de las etapas tempranas de la reproducción, técnicas de fertilización in vitro, expresión y caracterización de receptores de membranas plasmática, mecanismos de acción de agonistas, vías metabólicas, etc. El objetivo del presente trabajo con ovocitos de sapo Bufo arenarum es estudiar los mecanismos de acción de sustancias xenobióticas a nivel bioquímico y molecular, en relación con la capacidad de éstas gametas de ser fertilizadas y avanzar a través de los estadios tempranos del desarrollo embrionario. Se ha trabajado con diversos biomarcadores que permitieron reconocer los mecanismos de toxicidad de compuestos organoclorados y más recientemente de algunos metales pesados. Se demostró que la activación de una fosfolipasa C específica por el Dieldrin provoca depleción de mediadores lipídicos y como consecuencia inhibición de la fertilización (desarrollo hasta 4-8 células). Actualmente se estudian los efectos del Zn sobre la fertilización y su probable relación con alteraciones en la actividad de enzimas de la vía de las pentosas, directamente ligada a los mecanismos de detoxificación en presencia de stress oxidativo. El estudio de los mecanismos de citotoxicidad de diversas sustancias resulta de gran utilidad cuando se lleva a cabo en gametas, porque permite también evaluar los efectos de dichas sustancias sobre la reproducción de la especie. Los ovocitos de anfibios son además células de gran interés, por su utilidad en el estudio de la citotoxicidad de una amplia variedad de contaminantes hídricos

ABSTRACT

A proteolytic enzyme secreted by the first portion of amphibian oviduct, pars recta, called oviductin in Xenopus laevis, causes ultrastructural alterations on the extracellular matrix of coelomic eggs, turning them susceptible to fertilization. Although great advances have been made in the field of reproduction, the molecular mechanisms responsible for the fusion between the egg and the sperm are yet to be understood. We have recently demonstrated the presence of proteins from pars recta fluid in blood serum and extracellular matrix of coelomic eggs in Bufo arenarum. Here we show, using immunofluorescence procedures, that blood serum components are present in the extracellular matrix of coelomic and pars recta fluid-conditioned eggs in Bufo arenarum. Furthermore, by assessing the neutralizing effect on the conditioning activity of pars recta fluid on coelomic eggs we found that antibodies against pars recta secretions and blood serum inhibited the effect of sperm-lysin on the vitelline envelope of conditioned oocytes and impaired fertilization by sperm. Thus, serum proteins appear to be implicated in the molecular events that lead to amphibian fertilization

ABSTRACT

The present study describes the lung morphology of the common toad, Bufo arenarum, as observed by light and electron microscopy. The lung wall consists of four layers: mesothelium, dense connective tissue with thin elastic fibers, loose connective tissue containing smooth muscle fibers and an internal respiratory epithelium. The lung presents three types of folds defined by their epithelia. First order folds are coated by ciliated epithelium containing numerous goblet cells. Second order folds present the same type of epithelium but devoid of goblet cells, while third order folds are only lined by respiratory epithelium. The respiratory surface of the lung is lined by a single cell type, the pneumocyte, which presents characteristics of both type I and type II alveolar cells of higher vertebrates. The pneumocytes are prismatic in shape and possess attenuated cytoplasmatic processes which spread over the pulmonary capillaries to form the outer layer of the air-blood barrier. These cells present microvilli in the apical extreme and contain different types of cytoplasmic bodies: electron dense, multivesicular and lamellar

ABSTRACT

The aim of the present investigation was to standardize a method for measuring delta-aminolevulinic acid dehydratase (ALAD) activity in circulating red blood cells of adult Bufo arenarum kept in controlled environmental conditions, and to obtain reference basal values suitable for environmental monitoring of lead exposure. The normal ALAD activity for B. arenarum was 131.86 + 14.47 U per liter of red blood cells (n = 38, mean + SEM; interval 72.98 - 263.33). In animals exposed to lead, ALAD activity decreased as lead dose increased.

ABSTRACT

El plagicida organoclorado Dieldrin inhibe la fertilización de ovocitos de sapo, inseminados "in vitro" luego de 2 horas de incubación en solución de Ringer conteniendo 4 mg/l del pleguicida. La incubación con Dieldrin no conduce por sí misma a los ovocitos a la reacción cortical ni produce alteraciones evidentes en sus membranas vitelinas, pero sí favorece la formación de la membrana de fertilización cuando dichas células son inseminadas con posterioridad al tratamiento. La inhibición de la fertilización de ovocitos de sapo Bufo arenarum-Hensel por el Dieldrin, se produce como consecuencia de efectos del plaguicida sobre eventos posteriores a la reacción cortical, que da lugar a la formación de la membrana de fertilización

Se determinó la actividad de arginasa hepática en Bufo arenarum adulto luego de la administración de 2,8 y 5,0 mg Cd/Kg p.c., inyectados por tercios a lo largo de 26 días. La actividad específica de la enzima en los sapos inyectadod con Cd se redujo 37-55 por ciento con respecto a los controles. La reducción de la actividad podría ser considerada como respuesta bioquímica a cantidades subletales de Cd en una de las principales enzimas del ciclo de la urea

A simple technique for recording the EKG of the posterior lymphatic hearts of the toad Bufo arenarum (Hensel) in free moving unanesthetized specimens is described. This technique permits long term chronic recordings in varied physiological and behavioral conditions whereas it overcomes some of the technical difficulties encountered in obtaining reliable recordings.

ABSTRACT

We investigated the impact of environmental osmotic stress on the arginase activity if Bufo arenarum embryos. The activity at the first developmental stages was not sensitive to extreme variations of osmotic pressure of the environment. Later, at gill circulation, opercular fold and right operculum stages, the enzyme activity of embryos developed in concentrated solutions decreased significantly with respect to control. At complete operculum stage, the arginase activity increased 1.3-2.5 fold in all conditions, and was significantly higher in embryos grown in distilled water than in control animals

ANTECEDENTES: En un trabajo previo se demostró que un antagonista peptídico de la hormona antidiurética era capaz de bloquear el efecto de la angiotensina II en la permeabilidad al agua en presencia de un gradiente osmótico en la piel aislada del sapo, tejido con propiedades funcionales similares a las del nefrón de los mamíferos. En el presente trabajo se estudió el efecto de un antagonista de los receptores AT1 de la angiotensina II (Losartan) en la respuesta de la piel y vejiga aislada del sapo a la hormona antidiurética. MATERIAL Y METODO: La permeabilidad al agua en presencia de un gradiente osmótico fue medida por un método gravimétrico usando mitades simétricas de la piel ventral pélvica del sapo (Bufo arenarum). Después de mediciones controles se agregó Losartan al lado dérmico de una mitad, 10 minutos antes de una dosis de arginina vasotocina, la hormona antidiurética normalmente existente en los anfibios. Experimentos similares fueron realizados con vejigas urinarias usando una mitad como control y la otra como experimental. RESULTADOS: El Losartan no modificó la permeabilidad al agua en presencia de un gradiente osmótico basal en piel aislada de sapo pero inhibió significativamente la respuesta a la arginina vasotocina (10 menos 9M) en un 70,1 por ciento en concentraciones de 10 menos 5M, en 42,9 por ciento (10 menos 6M) y en 63,7 por ciento (10 menos 7M). El Losartan (10 menos 5M) no modificó en cambio la respuesta a la arginina vasotocina (10 menos 10M) en vejiga aislada de sapo. CONCLUSIONES: En la piel del sapo existiría un nuevo tipo de receptor común para la angiotensina II y la hormona antidiurética que difiere del receptor existente en la vejiga urinaria (V2), el cual sería exclusivo para la hormona antidiurética

ABSTRACT

Galectins (S-type or S-Lac lectins) are a well-defined family of beta-galactoside animal lectins characterized by a high sequence homology in the carbohydrate-binding domain. We have previously purified and characterized the S-type lectin from the ovary of the toad Bufo arenarum. In this study, we purified the S-type lectins from Bufo arenarum ovary and human spleen by an improved method which included ion exchange and affinity chromatography. Antibody cross-reactivities between both lectins and some other S-type lectins showed that they share epitopes. Glycosylation studies carried out with detection/differentiation kits suggested that both lectins are not glycosylated

ABSTRACT

The formation of vitelline envelope (VE) during the oogenesis of Bufo arenarum (Amphibia Anura) is described. At the stage of early vitellogenesis, the first structures appear: the number of oocyte microvilli increases, and many cross sections of them are observed between the follicle cells and the oocyte. A filamentous material is observed inside the follicle cells and between the follicle cells and the oocyte. Multivesicular bodies are also found in the follicle cells, and in the perivitelline space. The micrographs also suggest the participation of the oocyte in the process of VE formation: large vesicles are present in the cortex of the oocyte, filled with an amorphous material of low and uniform electron density. Some of them are in the process of releasing their content to the perivitelline space. Many vesicles (probably resulting from microvilli fragmentation) are also observed in the perivitelline space. During late vitellogenesis the VE is a continuous structure between the layer of follicle cells and the oocyte. The filamentous material is aggregated in bundles, forming a net, and the spherical components are now either included in the orifices of the net, or free near the oocyte's surface. At the end of oogenesis, when the VE is completely formed, it is difficult to distinguish its components independently. Immunolocalization with antibodies against VE, show a positive reaction in follicle cells and oocytes in previtellogenic and full grown ovarian follicles. This analysis suggests that both the oocyte and the follicle cells are directly involved in the synthesis and secretion of the components of the vitelline envelope in Bufo arenarum.
